题目: Conditional Cell Reprogramming for Living BioBank, Regenerative Medicine and Individualized Cancer Therapy


摘要:We describe a Conditional Reprogramming (CR) system which shows great promise for drug discovery, personalized cancer therapy and regenerative medicine. That is, combining Rho kinase inhibitor (Y-27632) and irradiated mouse fibroblast feeder cells induces normal and tumor epithelial cells to proliferate indefinitely in vitro.  The generation of cell cultures using this method is very rapid, robust and reproducible.  From a medical standpoint, it is significant that millions of cells can be generated from needle biopsies or cryopreserved tissue samples within a week and can generate cultures from many different types of tumors as well as normal tissue. The cells cultured from normal epithelium are morphologically undifferentiated and express adult stem cell markers, but can fully differentiate when placed into in vivo or in vitro conditions that mimic their natural environment. The induction of CR is rapid and results from reprogramming of the cell populations rather than a clonal selection as is the case with conventional cell lines. Unlike embryonic or induced pluripotent stem cells, conditionally reprogrammed cells (CRCs) from normal tissue do not express high levels Sox2, Oct4, Nanog, or Klf4 and do not form teratomas in xenografts.  Most important, CRCs do not need intricate manipulation to induce their differentiation into mature tissue. In addition, the technique has generated cultures from many different animals, including mouse, rat, ferret, pig, cow, and horse, etc.  In tumor cell cultures, the phenotypic and genotypic features are maintained between the primary tumor and the generated cultures and the technique has recently been used to identify an appropriate therapy for respiratory papillomatosis.  The applications for this method are far-reaching as it can be easily adapted for live biobanking, basic research, and diagnostic, therapeutic and regenerative medicine.


个人简介:张全顺 Quanshun ZHANG (美国体外科学研究院;高级科学家,教育和外联主管)  (IIVS; Senior Scientist, Director of Education and Outreach Programs) 现任体外科学研究院高级科学家以及教育和外联项目主任。体外科学研究院是非赢利的研究和检测实验室,致力于体外毒理学检测方法的验证,推广与应用。近年来与中检院,检验检疫局,CDC,北京工商大学,第4军大组织工程中心和中山大学等院校和企业建立了大量的合作项目。本人毕业于南京农业大学兽医(1987)和兽医微生物与免疫(1990)专业。曾任塔夫茨大学兽医学院比较医学的研究助理教授(负责HPLC和实验动物两个核心实验室)和上海动植物检疫局高级兽医师。获得过Pfizer Aspire 奖,农业部科技进步一等奖,国家动植检局的二等奖(1995)以及江苏省科技进步三等奖(1999)。其研究得到美国国立卫生研究院(RO1R21R56 NIH合同)以及辉瑞,MerckProgenicsMicrobiotix等的资助。发表论文30余篇。

Quanshun Zhang is currently a senior scientist and director of education and outreach programs at the Institute for In Vitro Sciences (IIVS). IIVS is a non-profit research and testing laboratory dedicated to the validation, adoption and application of in vitro (non-animal) methods worldwide. Founded in 1997, IIVS has worked with industry and government agencies to implement in vitro testing strategies that limit animal use while supplying key information for product safety and efficacy decisions. In China, IIVS has established wide cooperation with NIFDC/IFCC, AQSIQ/CAIQ/CIQ, CDC, Beijing Technology and Business University, the 4th Military Medical University Center for Tissue Engineering and Sun Yat-sen University. Quanshun Zhang graduated from Nanjing Agricultural University School of Veterinary Medicine, worked at Tufts University School of Veterinary Medicine as a research Assistant Professor (in charge of HPLC and Animal Core Facilities) and also worked at Shanghai Animal and Plant Quarantine Bureau as a Senior Veterinarian. His work was recognized by the following achievements: Pfizer Aspire Award, Ministry of Agriculture Science and Technology Advancement Award etc. His researches have been supported by Grants from National Institute of health (RO1, R21, R56 and the NIH contract), as well as by Funding from Pfizer, Merck, Progenics, Microbiotix etc. He published more than 30 scientific papers.
